Type A uncertainty เกิดจาก Random error ความไม่แน่นอนชนิดนี้หาค่าได้ด้วยการทำซ้ำ และประเมินด้วยวิธีทางสถิติ Type B uncertainty Systematic error ความไม่แน่นอนที่เกิดจากค่าที่อ่านได้จากเครื่องมือวัด หรือวัสดุที่ใช้ ไม่สามารถประเมินได้จากสถิติหรือการทำซ้ำ ประเมินได้จากการสอบเทียบเครื่องมือ หรือเอกสารรับรองวัสดุ เป็นต้น

38 การหาความไม่แน่นอน 1. “Bottom-up” Approach ตามวิธีของ GUM: 1993
พิจารณาทุกปัจจัยที่เกี่ยวข้องและมีผลต่อค่าการทดสอบ (แผนภูมิก้างปลา) และนำค่า uncertainty ของการวัดมารวมกัน แนวทางนี้มีรูปแบบที่ชัดเจนในโครงสร้าง และการกำหนดค่าความไม่แน่นอน นำไปสู่การพัฒนาทางเทคนิคการทดสอบ โดยใช้หลักการลด-เพิ่มค่าความไม่ แน่นอนในแต่ละปัจจัยได้

การประมาณค่าความไม่แน่นอนจากการนำค่าความผิดพลาดทั้งเชิง ระบบ และเชิงสุ่ม คือ Bias, repeatability และ reproducibility มา พิจารณาหาค่าความไม่แน่นอน ได้จากการทำซ้ำ และนำมาหาค่าเบี่ยงเบนมาตรฐานของความทำซ้ำได้ (Standard deviation of reproducibility, SR) ในทางจุลชีววิทยานิยมใช้ทางนี้ เนื่องจากปกติไม่สามารถประเมินค่า จริงได้ และ ค่าเบี่ยงเบนทางจุลชีววิทยาที่ยอมรับได้มีค่าสูง
